Tom Smith (3-68) checked the visitors' progress as they slipped to 140-4.
But Mitchell, who faced 228 balls and hit 13 fours, batted out the day - putting on 102 with Ben Cox (56) - and saw the visitors to 299-7 at the close.
Craig Miles (4-53) took the wicket of Shaaiq Choudhry (21) to leave the game finely poised after the first day's play.
BBC Hereford & Worcester's Dave Bradley:
"This was a good day's cricket. Daryl Mitchell was again the star for Worcestershire, a model of patience and consistency.
"Richard Oliver continues to impress, playing his shots with freedom, while Ben Cox continues his improvement, but Worcestershire lost three wickets post lunch to slow them down.
"What is a good score? Maybe if Worcestershire get to 350, they will think they have made it."
BBC Gloucestershire's Bob Hunt:
"Honours about even after day one.
"An interesting game on offer on a pitch that seemed to offer some assistance to the bowlers as the day progressed.
"It was good to see Craig Miles back for his first game of the season and his four-wicket haul was a just reward for bowling with venom throughout."
