Chris Cooke (52) and Jim Allenby (57) chipped in with half-centuries to help the visitors build a healthy lead.
Glamorgan eventually closed on 569-7, an advantage of 178, with Ruaidhri Smith adding an unbeaten 40.
BBC Radio Gloucestershire's Bob Hunt:
"A sparkling century from Ben Wright livened up things on day three, with Gloucestershire spending the day setting defensive fields and waiting for the batsmen to make mistakes.
"The home county will have to bat with the same discipline on the final day as they did over the final two days against Surrey in their previous match."
