Stephen Moore hit an unbeaten 106 as Derbyshire ended day two 203-2 on in reply to Hampshire's 328 all out.
The opener marched to his first century since arriving from Lancashire this winter, hitting 15 boundaries en route.
Resuming 231-5, Hampshire lost two more quickly, but Michael Bates (31) and Joe Gatting (67) helped post a solid total.
In reply, Billy Godleman departed early for Derbyshire, but Wayne Madsen made 36 and Shivnarine Chanderpaul (35 not out) stayed with Moore to stumps.
Having lost Will Smith for 70 late on day one, the responsibility at the start of play was on Gatting as Hampshire looking to move beyond 300.
And that looked a tough task when nightwatchman Kyle Abbott was clean bowled by Mark Footitt and Tim Groenewald had Sean Ervine caught behind soon after.
Gatting patiently made his way to a half-century before trying to raise the tempo, but holed out to long-on off Wes Durston to leave his side on 307-8, and Footitt then trapped Matt Coles lbw and Wes Durston had Bates caught to end the innings.
James Tomlinson trapped Godleman early in Derbyshire's reply, but Moore and Madsen rebuilt until the latter showed too much aggressive intent against Liam Dawson and was stumped.
Moore continued to bat calmly, and reached his 18th first-class hundred by depositing an Ervine delivery into the stands shortly before the close.
