Alan Richardson and Thilan Samaraweera produced superb individual performances as Worcestershire dominated the second day at Grace Road.
Seamer Richardson, 38, took the first hat-trick of his career to finish off the hosts' first innings for 274 after they had started the day on 270-7.
Samaraweera ended the day frustratingly unbeaten on 99 as the visitors recovered from 55-3 to close on 265-5.
He shared a fourth-wicket stand of 181 with Tom Fell, who hit 84.
Fell, 19, was caught behind off Shiv Thakor just 11 runs short of his best Championship score.
Richardson had taken two wickets on the first day and struck with the last ball of his 24th over and then the first two of his next with Ollie Freckingham, Matthew Hoggard and Alex Wyatt his victims.
