Gareth Roderick hit 152 not out to frustrate Kent as Gloucestershire closed on 307-9, chasing 399 to win.
Two quick declarations set-up a third-day run-chase but the visitors soon collapsed to 41-4 before lunch.
It was 138-6 when Roderick was joined by Tom Smith and the former marched to a maiden career century as they put on 109 before Charlie Shreck removed Smith and David Payne in quick succession.
But Gloucestershire stood firm and the game goes into a fourth day.
