Gloucestershire skittled Derbyshire for 95 and then built an 87-run first-innings lead on a busy day at Bristol.
The Division Two leaders looked relatively solid on 63-3 but then lost three wickets in 13 balls, including leading run-getter Wes Durston for 18.
There was no way back for the visitors as Will Gidman took 4-29 and fellow seamer David Payne 3-23.
In reply, Kane Williamson (56) and Alex Gidman (72 not out) led the home side to 182-5 at close.
Ross Whiteley (2-31) took two wickets in eight balls near the end of the day to get rid of Hamish Marshall (31) and Liam Norwell for one.
