Owais Shah scored his second century of the season but could not prevent Essex trailing going into the final day of their clash with Glamorgan in Cardiff.
After an hour's delay because of rain, Essex resumed on 160-2, but soon lost Jaik Mickleburgh (60).
But Shah went on to make 118, with Adam Wheater finishing unbeaten on 92 as Essex were dismissed for 363.
Alviro Petersen and William Bragg then both hit half-centuries as Glamorgan closed 162 ahead on 126-1.
Glamorgan's players wore black armbands in memory of former county and England all-rounder Allan Watkins, who died on Wednesday aged 89.
Players from both teams and 50 former Glamorgan players, who were having their annual lunch at the ground, observed a minute's silence during the lunch interval.
CLOSE OF PLAY SUMMARY
BBC Essex's Nick Gledhill:
"Essex chose not to declare behind and, thanks to the brilliance of Adam Wheater, were able to restrict their first innings deficit to 36.
"The game could then have drifted but the positive approach of Petersen and Bragg ensured that Glamorgan are on course for a lead above 280 by lunch on the final day.
"Victory for Glamorgan is probably essential if they are to stay in touch with the leaders so Essex will be hoping for a realistic target that gives both sides a chance."
