Chris Rogers led Middlesex's impressive reply with 87 after Gloucestershire were dismissed for 358 on day two.
Toby Roland-Jones took two more wickets to finish with 5-91 as Gloucestershire added 56 to their overnight 302-6.
Rogers then put on 116 for the first wicket with Scott Newman (47) before Jonathan Batty claimed three catches as Middlesex slipped to 194-3.
Dawid Malan (45) fell to Batty's fourth catch just before stumps as they closed on 296-4, Neil Dexter unbeaten on 56.

"If the first day belonged to Gloucestershire then the second was easily Middlesex's.
"Having allowed Gloucestershire to recover from 138-5 to 358 all out, Middlesex could have succumbed to scoreboard pressure in reply. But openers Chris Rogers and Scott Newman shared a fine century stand to set the foundation for the innings.
"Newman, along with Dan Housego and Dawid Malan, were all unfortunate to fall just short of their fifties. While Rogers produced an excellent 87 he will be disappointed not to have made a bigger score.
"The pitch looked even less helpful to the bowlers today but much will depend on skipper Neil Dexter on day three as he'll aim to guide the all-rounders to set a sizeable first innings lead."
