Durham take a lead of 264 runs into the third day after bowling out Sussex for 194 at Chester-le Street.
After starting day two on 30-1, in response to Durham's 292, only Andrew Hodd (67) and Chris Nash (65) made worthwhile runs for the visitors.
Callum Thorp took 3-52, backed by Ian Blackwell's 2-8 in 10.3 overs.
Blackwell then smashed a quickfire 51, hitting three sixes and six fours as Durham reached the close on 166-5.
Despite a first-innings lead of 98, Durham were rocking on 79-4 when Blackwell arrived at the crease.
But, after surviving a chance to slip off James Anyon on four and a return chance to Monty Panesar on five, Blackwell cashed in on his good fortune.
He hit Panesar just over the fielder at long-on, who was 10 yards in from the rope and that blew open the floodgates.
Three sixes followed, two of them fiercely pulled in one Anyon over, he reached 50 off 44 balls.
But, two balls later, he chanced his arm once too often, holing out to deep square leg.

"Good performances from Nash and Hodd, but sadly an all too familiar Sussex batting collapse.
"Conditions changed when Durham came out to bat again, the mist started to roll in slowly and the ball started to swing, with my personal pick of the bowlers Rana Naveed making it move appreciably. But James Anyon was carted to the boundary by Blackwell for 16 in three balls.
"Sussex will now need to post their biggest score of the season to win, or bat nearly a day and a half to draw. But it looks very likely that we will see a completion of this match tomorrow."
