Cheteshwar Pujara scored his second successive half-century in India's final tour match before the first Test against England.
Derbyshire declared on their overnight 326-5 and Pujara, who hit 57 against Leicestershire last week, scored 81 before retiring out.
All-rounder Stuart Binny made 81 not out as India declared on 341-6.
The first Test of the five-match series starts on Wednesday, 9 July at Trent Bridge.
India's first tour game against Leicestershire ended in a draw and Thursday's last day at Derby will see the 90 overs split between both teams' second innings.
The tourists made a poor start to the second day as they were reduced to 18-2, with openers Murali Vijay and Shikhar Dhawan dismissed cheaply.
But Pujara, who averages 58 in 19 Tests, repaired the damage with Virat Kohli (36) and India captain MS Dhoni (46).
Pujara is yet to be dismissed in England this summer after retiring against Leicestershire.
Ravindra Jadeja, dropped at slip by Chesney Hughes off Greg Cork on nought, made 45 before he was the last man out.
