Simon Kerrigan took 5-68 and three Lancashire batsmen hit fifties to set the the hosts up for a final-day declaration against Gloucestershire.
The visitors began on 143-4, before the spinner took five of the six wickets.
Centurion Alex Gidman (110) was his final victim as Gloucestershire were eventually dismissed for 222.
Ashwell Prince, Gareth Cross (both 64) and 52 not out from Wayne White led Lancashire to 239-7 at stumps, an overnight lead of 347.

"An excellent day for Lancashire who find themselves in charge of the game heading into the final day.
"Simon Kerrigan took 4-40 in 14 overs this morning to bowl Gloucestershire out and secure a first-innings lead of 88 runs.
"The only question for Lancashire now is when to declare. They need to give themselves time to take 10 wickets on the final day to clinch their third Championship win of the season."

"Gidman's innings was superb under the circumstances and the attitude of Ed Young, who came in at number 11 with his wrist in plaster, epitomised the fighting qualities that John Bracewell wants to install in this young side.
"Young assisted Gidman in reaching a batting bonus point and then oversaw him reaching his hundred.
"With Young's injury leaving Gloucestershire a bowler light, it was always going to be a hard toil for a youthful attack.
"But they battled hard and will be looking to clear up the Lancashire tail in the morning."
