Alex Gidman's first century for over a year helped Gloucestershire build a significant lead before Derbyshire fought back on day two at Bristol.
Resuming on 182-5, Gidman added 55 to his overnight total to reach 129 before the hosts were bowled out for 294.
Having been reduced to 15-1 in their second innings, opener Martin Guptill hit 42 to steady Derbyshire's nerves.
Wayne Madsen (72) and Dan Redfern (64 no) helped the visitors reach 209-5 at the close, a lead of 10 runs.
