Tesco is cutting 150 jobs at its distribution centre at Magor in Monmouthshire.
The supermarket group said: "We've been reviewing our distribution network, and have today confirmed plans for a slight reduction in our Magor operation."
The company employs 800 staff at the site.
The Usdaw union, which represents the workers, said the losses are "devastating".
Workers will be offered jobs at other locations and new distribution centres are to open in Reading and Dagenham.
A Tesco spokesman said staff have been informed and the company was "working with them and their representatives".
Usdaw national officer Joanne McGuinness said: "This is absolutely devastating news for our members and their families.
"We will be entering into a 90 day consultation with Tesco to explore all possible options to reduce any job losses."
